目的探讨糖尿病患者外周血单个核细胞(PBMCs)中血管内皮生长因子(VEGF)基因表达变化,分析其与糖尿病肾病进展的相关性。方法选取本院内分泌科2013年-2014年51例糖尿病患者作为实验组,50例健康体检者作为对照组,real-time PCR法检测糖尿病患者PBMCs VEGF基因mRNA表达水平,酶联免疫吸附试验(ELISA)测定血清中VEGF水平。结果与对照组比较,糖尿病组VEGF基因mRNA表达升高,血清VEGF水平也升高,差异有统计学意义(P<0.05)。糖尿病PBMCs VEGF基因mRNA表达量随着糖尿病肾病严重程度依次升高:DN3>DN2>DN1>DN0,而DN3组、DN2组血浆VEGF水平显著高于DN1组、DN0组,差异有统计学意义(P<0.05)。结论糖尿病患者PBMCs VEGF基因表达显著升高,与糖尿病肾病发生发展呈正相关,或可成为监测糖尿病肾病进展又一靶标。
Objective To investigate the changes of vascular endothelial growth factor (VEGF) gene expression in peripheral blood mononuclear cells (PBMCs) of diabetic patients and its relationship with the progression of diabetic nephropathy. Methods Fifty-one patients with diabetes mellitus from 2013 to 2014 were selected as the experimental group and 50 healthy subjects as the control group. Real-time PCR was used to detect the mRNA expression of VEGF gene in PBMCs of diabetic patients. Enzyme-linked immunosorbent assay (ELISA) ) To measure serum VEGF levels. Results Compared with the control group, the mRNA expression of VEGF in diabetic group increased and the serum VEGF level also increased, the difference was statistically significant (P <0.05). The expression of VEGF mRNA in diabetic PBMCs increased with the severity of diabetic nephropathy: DN3> DN2> DN1> DN0, while the levels of VEGF in DN3 and DN2 groups were significantly higher than those in DN1 and DN0 groups (P < <0.05). Conclusion The expression of VEGF gene in PBMCs of diabetic patients is significantly increased, which is positively correlated with the occurrence and development of diabetic nephropathy. It may be another target of monitoring the progression of diabetic nephropathy.
