应用RP-HPLC方法检测血清中氟脲嘧啶(Fluorouracil)浓度,本方法采用C18分析柱及预柱,流动相为甲醇-0.5%乙酸水溶液(5:95v/v),检测波长266nm,血清样品用乙酸乙酯萃取,氮气吹干,血药浓度在25ng~40μg/ml范围内呈线性关系,相关系数0.9999,最低检测浓度为25ng/ml。生物样品的方法回收率为101.2%~107.7%,日内日间变异系数在1.13%~6.24%(n=5)。
The concentration of Fluorouracil in serum was determined by RP-HPLC method. The method was based on C18 column and precolumn. The mobile phase consisted of methanol-0.5% acetic acid aqueous solution (5:95 v / v) The sample was extracted with ethyl acetate and dried under nitrogen. The linear range was 25 ng ~ 40 μg / ml with a correlation coefficient of 0.9999 and the lowest detection concentration was 25 ng / ml. The recoveries of biological samples ranged from 101.2% to 107.7%, and the intra-day and intra-day CVs ranged from 1.13% to 6.24% (n = 5).
