目的 测定原发性肝癌病人肝动脉化疗栓塞后细胞凋亡、端粒酶活性、p53蛋白、sIL 2R和T淋巴细胞亚群表达的变化 ,进而比较不同药物联合TACE的治疗效果。方法 60例原发性肝癌病人 ,按治疗药物不同分 4组 ,用TUNEL法、TRAP PCR ELISA法及免疫组化法 ,分别测定细胞凋亡、端粒酶活性、T淋巴细胞亚群及sIL 2R的表达。结果 60例HCC细胞凋亡、端粒酶和p53蛋白阳性率均明显高于对照组 (P <0 0 1) ,p53蛋白阳性率与AI呈负相关 (r =-0 42 3 ,P <0 0 5) ;治疗后sIL 2R的表达较治疗前下降 (P <0 0 5) ,CD+ 3 、CD+ 4和CD+ 3 /CD+ 4比值较治疗前上升 ,并与疗效有关 ;TACE +PEI组和TACE +LAK组疗效均高于常规组 (P <0 0 5)。结论 TACE +PEI组和TACE +LAK组治疗原发性肝癌效果较好
Objective To determine the changes of apoptosis, telomerase activity, p53 protein, sIL 2R and T lymphocyte subsets in patients with primary liver cancer after chemoembolization, and then compare the therapeutic effects of different drugs combined with TACE. Methods Sixty patients with primary hepatocellular carcinoma (HCC) were divided into 4 groups according to different therapeutic agents. TUNEL, TRAP PCR and immunohistochemistry were used to detect the apoptosis, telomerase activity, T lymphocyte subsets and sIL 2R expression. Results The positive rate of telomerase and p53 protein in 60 HCC cells was significantly higher than that in control group (P <0.01). The positive rate of p53 protein was negatively correlated with AI (r = -0 423, P <0 (P <0.05). The ratio of CD + 3, CD + 4 and CD + 3 / CD + 4 in sIL 2R was significantly higher than before treatment (P <0.05) + LAK group were higher than the conventional group (P <0 05). Conclusions TACE + PEI group and TACE + LAK group have better effect on primary liver cancer
