杂交子代的纯度直接影响到遗传图谱的质量,因而在遗传图谱构建前要对杂交子代进行鉴定与选择。笔者从300对SSR标记中,筛选出11对能明确显示亲本来源的标记,利用这11对标记对380株美洲黑杨杂交子代进行了鉴定。结果表明:有372个个体因同时具有亲本各1条等位基因而被认定为亲子代可用于图谱构建,其余8个个体因产生了非亲本来源或额外的等位基因,这些个体可能源于花粉污染或染色体异常,在作图过程中宜从作图群体中排除。
The purity of the hybrid progeny directly affects the quality of the genetic map, and therefore the hybrid progeny should be identified and selected before the genetic map is constructed. From 300 pairs of SSR markers, the author screened 11 pairs of markers that can clearly show the origin of the parents, and used the 11 pairs of markers to identify 380 hybrid poplars of Populus nigra. The results showed that 372 individuals were identified as parental offspring by having one allele each at the same time and could be used to construct the map. The remaining eight individuals were derived from non-parental origin or additional alleles. These individuals may be derived from Pollen or chromosomal abnormalities should be excluded from the mapping population during the mapping process.
