目的:寻找一种快速高效的口腔软硬组织联合切片方法。方法:选择30只大鼠取出带有牙齿的上颌骨,经组织固定后随机分成A、B、C组,每组10个标本,以10%EDTA脱钙液在不同条件下脱钙,经HE制片观察其染色情况,判定脱钙效果。结果:A组室温静置脱钙时间为(50±6)d,B组室温静置脱钙基础上,每天用37℃搅拌器搅拌4 h,脱钙时间为(28±2)d,C组室温静置脱钙基础上,每天用37℃搅拌器搅拌4 h及超声波处理4 h,脱钙时间为(10±3)d,3组标本HE染色差异无统计学意义。结论:EDTA恒温搅拌加超声波联合脱钙方法在牙及牙周组织联合切片中能够大大缩短脱钙时间,并有良好的制片效果。
Objective: To find a fast and efficient oral and soft tissue joint biopsy method. METHODS: Thirty rats were selected to remove the maxilla with teeth. After fixation, the rats were randomly divided into groups A, B and C, 10 specimens in each group were decalcified with 10% EDTA decalcification solution under different conditions. Results: The time of decalcification in group A was (50 ± 6) d at room temperature, the decalcification time was (28 ± 2) d in group B at 37 ℃ for 4 h, On the basis of the group of decalcified at room temperature, the decalcification time was (10 ± 3) d with stirring at 37 ℃ for 4 h and sonication for 4 h. There was no significant difference in HE staining between the three groups. CONCLUSION: EDTA constant temperature stirring combined with ultrasonic decalcification method can greatly shorten the decalcification time in the combination of teeth and periodontal tissues, and has a good film production effect.
