采用超临界CO_2流体萃取技术研究荷叶总生物碱的提取工艺条件。以萃取温度、萃取压力、夹带剂流速、萃取时间为工艺参数,通过单因素试验和正交试验相结合的方法,确定超临界CO_2流体萃取荷叶总生物碱的最佳工艺,即:萃取温度55℃,萃取压力20MPa,夹带剂流速0.2mL/min,萃取时间2h。各因素影响荷叶总生物碱得率的次序为:萃取温度>萃取时间>萃取压力>夹带剂流速。最佳工艺验证试验的荷叶总生物碱得率为318.45μg/g。
Study on Extraction Technology of Total Alkaloids in Lotus Leaf by Supercritical CO2 Fluid Extraction. The optimal extraction process of total alkaloids from lotus leaf by supercritical CO 2 fluid was determined by single factor experiment and orthogonal experiment with extraction temperature, extraction pressure, entrainer flow rate and extraction time as process parameters, ie extraction temperature 55 ℃, extraction pressure 20MPa, entrainer flow rate 0.2mL / min, extraction time 2h. The factors influencing the yield of total alkaloids in lotus leaf were: extraction temperature> extraction time> extraction pressure> entrainer flow rate. The best process validation test lotus leaf total alkaloid yield was 318.45μg / g.
