[Objective]To study protective effect and acting mechanism of total flavonoids of POLYGONI PERFOLIATI HERBA on acute alcohol-induced liver injury in mice. [Methods]60 mice were randomly divided into normal group,model group,Bifendate positive group(150 mg / kg),and high( 600 mg / kg),medium( 400 mg / kg) and low( 200 mg / kg) dosage groups of PPTF,with 10 mice for each group. Except the normal group,fill the stomach with 12 mL/kg 56% liquor one time every morning for the rest groups and fill the corresponding dosage liquor in the afternoon,and fill the same volume of distilled water for normal group and model group. Continuously implement these operations for 14 days. After the end of the last time stomach filling,take fasting for 24 hours,anaesthetize them and take blood,and collect livers; measure the activity or content of alanine transaminase( ALT),aspartate transaminase( AST),malondialdehyde( MDA),and superoxide dismutase( SOD) in serum by biochemical method; measure the content of interleukin-4( IL-4),interleukin-6( IL-6),tumor necrosis factor-α( TNF-α) in liver tissue by enzyme-linked immunosorbent assay( ELISA) method; HE dyeing,and observe pathological changes of liver tissue by a microscope. [Results] Compared with the model group,all dosage groups of total flavonoids of POLYGONI PERFOLIATI HERBA can effectively reduce activity and content of ALT,AST and MDA in serum of mice,can improve SOD activity,and can reduce IL-6 and TNF-α in liver tissue,and increase IL-4 content. [Conclusions] Total flavonoids of POLYGONI PERFOLIATI HERBA have protective effect on acute alcohol-induced liver injury in mice and the mechanism is possibly related to anti-lipid peroxidation and inhibition of inflammation reaction in liver.
