目的研究肝细胞肝癌(HCC)组织中Spy1的表达,探讨其与增殖细胞核抗原(PCNA、Ki67)以及与临床病理之间的关系。方法免疫组化方法检测50例HCC及癌旁组织和10例肝血管瘤旁肝组织中Spy1和Ki67蛋白的表达。收集5例HCC及癌旁肝新鲜组织,免疫印迹技术检测Spy1和PCNA的表达。结果Spy1在HCC中的表达高于对应的癌旁肝组织。HCC中,Spy1的表达强度与肿瘤病理分级?血清AFP值之间相关(P<0.05);Spy1与Ki67表达呈正相关(P<0.01)。结论Spy1在HCC组织中高表达,Spy1可能在HCC的发生、发展中发挥重要的作用。
Objective To investigate the expression of Spy1 in hepatocellular carcinoma (HCC) tissues and to explore the relationship between the expression of Spy1 and the expression of proliferating cell nuclear antigen (PCNA, Ki67) and its clinical pathology. Methods Immunohistochemistry was used to detect the expression of Spy1 and Ki67 in 50 cases of HCC, adjacent non-cancerous tissues and 10 cases of hepatic hemangiomas. Five cases of HCC and paracancer liver tissues were collected and the expression of Spy1 and PCNA was detected by Western blotting. Results The expression of Spy1 in HCC was higher than that in corresponding paracancer liver tissue. In HCC, the expression of Spy1 was correlated with tumor grade and serum AFP (P <0.05), but there was a positive correlation between Spy1 and Ki67 (P <0.01). Conclusion Spy1 is highly expressed in HCC tissues. Spy1 may play an important role in the development of HCC.
