目的:观察参芪补肺汤对慢性阻塞性肺疾病(COPD)肺气虚证大鼠肺细胞外基胶原和纤维连接蛋白(FN)的影响。方法:用烟、SO2定量熏吸并复合木瓜蛋白酶雾化吸入法,建立COPD肺气虚证大鼠模型;60只雄性Wistar大鼠随机分为正常组、模型组、参芪补肺汤高、中、低剂量组、激素组;肺Masson染色观察胶原纤维的分布情况;放射免疫法检测支气管肺泡灌洗液(BALF)中FN的含量。结果:与正常组比较,模型组支气管肺组织单位面积内胶原面积和积分吸光度(IA)显著增加(P≤0.01);BALF中FN的含量显著增加(P<0.01)。经治疗后,与模型组比较,参芪补肺汤高、中剂量组胶原面积和积分吸光度(IA)显著降低(P<0.05);FN含量显著减少(P<0.01)。结论:参芪补肺汤可能通过抑制COPD肺气虚证大鼠肺细胞外基质的过度沉积,而对阻止COPD气道重构有一定作用。
Objective: To observe the effect of Shenqibufei decoction on extracellular lung collagen and fibronectin (FN) in rats with chronic obstructive pulmonary disease (COPD). Methods: The COPD model of lung qi deficiency syndrome was established by inhalation of cigarette smoke and SO2, and the rats were randomly divided into normal group, model group, Shenqi Bufei Tang, , Low dose group and hormone group. Masson staining of lungs was used to observe the distribution of collagen fibers. Radioimmunoassay was used to detect the content of FN in bronchoalveolar lavage fluid (BALF). Results: Compared with the normal group, the collagen area and integral absorbance (IA) of bronchial lung tissue in model group increased significantly (P≤0.01). The content of FN in BALF increased significantly (P <0.01). After treatment, compared with the model group, Shenqi Bufei Decoction high and middle dose group collagen area and integral absorbance (IA) was significantly lower (P <0.05); FN content was significantly reduced (P <0.01). Conclusion: Shenqi Bufei Decoction may play an important role in preventing airway remodeling of COPD by inhibiting the excessive deposition of extracellular matrix of lung in rats with COPD.
