目的研究白藜芦醇促进斑马鱼尾鳍再生的活性及作用机制,并研究其对斑马鱼胚胎发育的影响,为白藜芦醇药效的进一步开发和安全用药提供依据。方法采用斑马鱼(Fli1-GFP)模型,研究白藜芦醇对斑马鱼尾鳍切除后血管和组织再生的影响,用相对荧光定量PCR法检测仔鱼切尾后体内血管内皮生长因子(VEGFA)和受体(VEGFR2)基因的表达量;研究白藜芦醇对斑马鱼胚胎发育的影响,检测其对斑马鱼胚胎自主运动、心率、孵化率的效应。结果白藜芦醇具有血管生成活性,可促进斑马鱼尾鳍再生,能提高仔鱼切尾后体内VEGFA及VEGFR2基因的表达量;白藜芦醇对斑马鱼胚胎发育有一定影响,主要表现为低浓度时促进自主运动和孵化率但高浓度时呈抑制效应,且可降低心率。结论白藜芦醇可促进斑马鱼尾鳍再生,作用机制可能与促进VEGFA和VEGFR2基因表达有关。白藜芦醇对斑马鱼胚胎发育有一定毒性。
OBJECTIVE: To study the activity and mechanism of resveratrol on the regeneration of zebrafish caudal fin, and to study the effect of resveratrol on the development of zebrafish embryo, so as to provide evidence for the further development and safety of resveratrol. Methods The effects of resveratrol on the regeneration of blood vessels and tissues after tail fin excision were studied by using zebrafish (Fli1-GFP) model. The relative fluorescence quantitative PCR was used to detect the changes of vascular endothelial growth factor (VEGFA) (VEGFR2) gene expression; study Resveratrol on zebrafish embryonic development, to detect their effects on zebrafish embryo autonomy, heart rate, hatchability. Results Resveratrol had angiogenic activity, which could promote the regeneration of zebrafish caudal fin, and increase the expression of VEGFA and VEGFR2 gene in the tail of larval fish. Resveratrol had some effects on the embryo development of zebrafish, When to promote voluntary movement and hatching rate, but when the concentration was inhibited, and can reduce heart rate. Conclusion Resveratrol can promote the regeneration of zebrafish caudal fin, the mechanism may be related to the promotion of VEGFA and VEGFR2 gene expression. Resveratrol has certain toxicity on zebrafish embryo development.
