目的分析系统性红斑狼疮(SLE)患者外周血中调节性T细胞(Treg)表达情况,初步探讨其在SLE患者细胞免疫失调中的意义。方法采用流式细胞术检测50例SLE患者和31例健康人对照组外周血中CD4+CD25+CD127low/-Treg细胞百分率,并分析其与CD4+CD25+和CD4+CD25high细胞的相关性,以及各种Treg细胞与SLE患者及SLE疾病活动性指数(SLEDAI)的相关性。结果 SLE活动期外周血CD4+CD25high细胞百分数为(11.65±5.53)%,显著高于SLE稳定期的(8.52±3.15)%和健康人对照组的(6.85±1.63)%,F=23.287,P<0.01。SLE患者Treg细胞与CD4+CD25high和CD4+CD25+细胞百分数均呈正相关(r分别为0.819,0.711,P均<0.05),SLEDAI与CD4+CD25+CD127low/-、CD4+CD25+、CD4+CD25high细胞水平呈正相关(r分别为0.410,0.394,0.348,P均<0.05)。结论 SLE活动期外周血中Treg细胞表达增加,且与SLEDAI相关,SLE患者免疫异常可能与Treg细胞的异常水平有关。
Objective To analyze the expression of regulatory T cells (Tregs) in peripheral blood of patients with systemic lupus erythematosus (SLE) and to explore its significance in cellular immune disorders of SLE patients. Methods Flow cytometry was used to detect the percentage of CD4 + CD25 + CD127low / -Treg cells in peripheral blood of 50 SLE patients and 31 healthy controls. The correlations between CD4 + CD25 + and CD4 + CD25high cells were analyzed, Treg cells and SLE patients and SLE disease activity index (SLEDAI) correlation. Results The percentage of CD4 + CD25high cells in peripheral blood of SLE patients was (11.65 ± 5.53)%, which was significantly higher than that of healthy controls (8.52 ± 3.15%) and healthy subjects (6.85 ± 1.63)%, F = 23.287 <0.01. The percentage of Treg cells in CD4 + CD25high and CD4 + CD25 + cells in SLE patients were positively correlated (r = 0.819, 0.711, P <0.05, respectively) (R = 0.410,0.394and0.348 respectively, all P <0.05). Conclusion The expression of Treg cells in the peripheral blood of patients with SLE increased, and associated with SLEDAI. The abnormality of SLE patients may be related to the abnormal level of Treg cells.
