目的:探讨夏枯草提取物对急性心力衰竭(AHF)模型大鼠心肌保护作用及基质细胞衍生因子-1(SDF-1)表达的影响,探讨其治疗AHF的可能作用机制。方法:采用Wistar大鼠建立AHF模型,随机分为4组,AHF模型组(AHF组),给予等体积生理盐水ig;夏枯草低剂量组,按照夏枯草提取物0.5 g·kg-1剂量ig;夏枯草中剂量组,按照夏枯草提取物1.0 g·kg-1剂量ig;夏枯草低剂量组,按照夏枯草提取物2.0 g·kg-1剂量ig;另设假手术组(Sham组),给予等体积生理盐水ig。6周后进行血流动力学、超声心动图测量,并分别采用流式细胞术,苏木素-伊红(HE)染色测量CD34+细胞阳性率,微血管密度(MVD)。进一步采用蛋白质免疫印迹(Western blot)和实时荧光定量-聚合酶链式反应(q PCR)分析检测SDF-1蛋白和SDF-1mRNA表达的变化。结果:夏枯草提取物可有效改善AHF大鼠左心室舒张功能,减轻心室壁厚度,促进CD34+细胞的动员和新生血管的形成,与AHF组比较,差异有统计学意义(P<0.05,P<0.01)。夏枯草提取物可使SDF-1蛋白和SDF-1 mRNA的表达显著增加,与AHF组比较,差异有统计学意义(P<0.05,P<0.01)。结论:夏枯草提取物对衰竭心脏有明显的保护作用,其机制可能与促进新生血管的形成,升高SDF-1表达水平和动员CD34+细胞释放至外周血有关。
Objective: To investigate the effects of Prunella vulgaris extract on myocardial protection and the expression of stromal cell-derived factor-1 (SDF-1) in rats with acute heart failure (AHF) and to explore its possible mechanism of action in treating AHF. Methods: AHF model was established in Wistar rats and randomly divided into 4 groups, AHF model group (AHF group), given an equal volume of normal saline ig; Prunella low dose group, according to Prunella vulgaris extract 0.5 g · kg -1 dose ig ; Prunella medium dose group, according to Prunella extract 1.0 g · kg-1 dose ig; Prunella low dose group, according to Prunella vulgaris extract 2.0 g · kg-1 dose ig; another sham group (Sham group) , Given an equal volume of saline ig. Six weeks later, hemodynamic and echocardiographic measurements were performed. The positive rate of CD34 + cells and the microvessel density (MVD) were measured by flow cytometry and hematoxylin-eosin staining respectively. The changes of SDF-1 protein and SDF-1 mRNA expression were further detected by Western blot and q-PCR. Results: Prunella vulgaris extract could effectively improve left ventricular diastolic function, reduce ventricular wall thickness, promote CD34 + cell mobilization and neovascularization in AHF rats. Compared with AHF group, the extract of Prunella vulgaris showed significant difference (P <0.05, P < 0.01). Prunella vulgaris extract could significantly increase the expression of SDF-1 protein and SDF-1 mRNA, compared with AHF group, the difference was statistically significant (P <0.05, P <0.01). CONCLUSION: Prunella vulgaris extract has obvious protective effect on failing heart. Its mechanism may be related to promoting the formation of new blood vessel, increasing the expression of SDF-1 and mobilizing CD34 + cells to release into peripheral blood.
