目的 :研究急性白血病 (AL)细胞BCL 2表达及其与体外化疗药物敏感性的关系。方法 :采用Westernblotting技术和四甲基偶氮唑盐比色法 (MTT法 ) ,分别检测HL 60、U93 7细胞株和 6例急性髓性细胞白血病 (AML)细胞BCL 2表达水平及体外对柔红霉素 (DNR)的敏感性。结果 :( 1)BCL 2表达水平HL 60细胞高于U93 7细胞 ,而 6例AML细胞BCL 2表达水平不同 ;( 2 )MTT检测结果显示 ,DNR对HL 60抑制率低于U93 7;BCL 2高表达的AML细胞 ,DNR抑制率低。结论 :细胞系和白血病原代细胞的BCL 2高表达伴低药敏与AML疗效有关。
Objective: To investigate the expression of BCL 2 in acute leukemia (AL) cells and its relationship with chemosensitivity in vitro. Methods: Western blotting and MTT assay were used to detect the expression of BCL 2 in HL 60, U93 7 cells and 6 cases of acute myeloid leukemia (AML) cells, respectively. Erythromycin (DNR) sensitivity. Results: (1) The expression of BCL 2 in HL 60 cells was higher than that in U93 7 cells, while the expression levels of BCL 2 in 6 cases were different. (2) The MTT assay showed that the inhibition rate of DNR to HL 60 was lower than that of U93 7; High expression of AML cells, DNR low inhibition rate. CONCLUSION: High expression of BCL2 in primary cell lines and primary leukemia cells with low susceptibility is associated with the efficacy of AML.
