多种细胞因子参与了哮喘时嗜酸性粒细胞与血管内皮细胞粘附的调节,但对嗜酸性粒细胞具有选择性作用的细胞因子尚未澄清。采用双色免疫荧光流式细胞技术,研究了炎前性细胞因子TNFα对人外周血嗜酸性粒细胞表面具有介导细胞粘附作用的粘附分子L-选择素和极迟抗原-4表达的影响,并与嗜中性粒细胞作以比较。TNFα对嗜酸性粒细胞和嗜中性粒细胞表面L-选择素均有上调表达的作用,而对极迟抗原-4表达无明显影响。实验结果说明,TNFα对L-选择素介导的嗜酸性粒细胞粘附无选择性作用。
A variety of cytokines are involved in the regulation of eosinophil adhesion to vascular endothelial cells in asthma, but cytokines selective for eosinophils have not yet been clarified. The effects of proinflammatory cytokines TNFα on the expression of adhesion molecule L-selectin and the expression of very late antigen-4 with mediating cell adhesion on the surface of human peripheral blood eosinophils were studied by two-color immunofluorescence flow cytometry , And compared with neutrophils. TNFα up-regulated the expression of L-selectin on eosinophils and neutrophils, but had no significant effect on the expression of late-stage antigen-4. The experimental results indicate that TNFα has no selective effect on L-selectin-mediated eosinophil adhesion.
