目的以鸡、猪等动物肝脏为研究对象,二氯甲烷为提取溶剂,建立免疫亲和柱净化、高效液相色谱-光化学衍生-荧光检测器检测六种黄曲霉毒素的分析方法。方法样品经提取、过滤、浓缩、复溶后经免疫亲和柱净化,进样后采用光化学衍生器在线对黄曲霉毒素G1、B1衍生,荧光检测器分析B1、B2、G1、G2、M1、M2六种黄曲霉毒素。结果在不同加标水平,不同肝脏回收率在70.2%~94.1%之间,变异系数为2.67%~12.90%,检测低限为0.16μg/kg。结论该方法的准确度与精密度较高,可以满足痕量分析要求。
OBJECTIVE To establish a method for the determination of six aflatoxins by immunoaffinity column purification and high performance liquid chromatography-photochemical derivatization-fluorescence detector using the liver of chickens and pigs as the research object and methylene chloride as the extraction solvent. Methods The samples were extracted, filtered, concentrated, reconstituted and purified by immunoaffinity column. After injection, aflatoxin G1 and B1 were derivatized by photochemical derivatization. The fluorescence intensity of B1, B2, G1, G2, M1, M2 six aflatoxins. Results At different spiked levels, the recoveries of different liver ranged from 70.2% to 94.1% with a coefficient of variation of 2.67% to 12.90% and a detection limit of 0.16μg / kg. Conclusion The method has high accuracy and precision, which can meet the requirements of trace analysis.
