用包埋脱水法冷冻保存水稻胚性悬浮细胞。整个过程包括:胚性悬浮细胞预培养、细胞包埋、二次预培养、包埋细胞脱水、液氮冰冻、细胞解冻和冷冻细胞恢复培养。结果表明,在细胞水分含量为25.17%和蔗糖浓度依次递增以及第2次预培养3~4d的存活率最好。在培养基中加2.5g·L-1活性炭有利于细胞的恢复生长。细胞恢复培养后,能再产生愈伤组织,但生长变慢,有约5d的滞后期。
Cryopreservation of Rice Embryogenic Suspension Cells by Embedding Dehydration. The whole process includes pre-culture of embryogenic suspension cells, cell embedding, secondary pre-culture, embedded cell dehydration, liquid nitrogen freezing, cell thawing and frozen cell recovery culture. The results showed that the cell survival rate was the best when cell moisture content was 25.17%, sucrose concentration was increased in sequence, and the second pre-culture was 3-4 days. Add 2.5g · L-1 activated carbon in the medium is conducive to cell growth and recovery. After the cells were recovered and cultured, the callus could be regenerated, but the growth slowed down with a lag of about 5 days.
