目的探索分解卵巢黏液性癌表面黏液的方法及其对化疗药物泰素作用于人卵巢黏液性癌细胞株OMC685的影响。方法2007年4月至11月于复旦大学妇产科研究所运用免疫细胞化学法分析OMC685所分泌黏液中粘蛋白的类型,通过查阅文献筛选出适当的黏蛋白分解酶,用ELISA法证明黏液的分解,并于黏液分解后加用泰素,MTT法检测OMC685增殖的变化。选取人卵巢浆液性癌细胞株SKOV3加入含黏蛋白培养液后,再给予泰素,MTT法检测细胞增殖。结果OMC685表面所包绕黏液成分为黏蛋白2(mucin2)。α-N-内切乙酰半乳糖苷酶能够较特异地分解黏液。黏液分解后加用泰素对OMC685细胞增殖的抑制显著增强,而SKOV3加含粘蛋白的培养液后再加入泰素,对细胞增殖的抑制作用显著减弱。结论卵巢黏液性癌细胞分泌的黏液可能起到一定的屏障保护作用,通过使用分解酶分解黏液能够提高化疗药物泰素的作用效果。
Objective To explore the method of decomposing mucus in ovarian mucinous carcinoma and its effect on the chemotherapeutic drug taxol in human ovarian mucinous carcinoma cell line OMC685. Methods Immunofluorescence was used to analyze the type of mucin in mucus secreted by OMC685 from April 2007 to November in Fudan University Institute of Obstetrics and Gynecology, Fudan University. The appropriate mucolytic enzyme was screened out by literature review. The mucinous Decomposition, and in the mucoid decomposition plus Taxol, MTT assay OMC685 proliferation changes. Human ovarian serous carcinoma cell line SKOV3 was added into mucinous medium and then paclitaxel was added. Cell proliferation was detected by MTT assay. Results OMC685 surface mucin is surrounded by mucin 2 (mucin2). Alpha-N-endo-acetylgalactosidase is able to break down mucus more specifically. Inhibition of proliferation of OMC685 cells by mucolytic treatment with Taxol was significantly enhanced, while the addition of paclitaxel to SKOV3 plus mucin-containing medium significantly reduced the inhibition of cell proliferation. Conclusion The mucus secreted by ovarian mucinous carcinoma cells may play a role in barrier protection. The action of paclitaxel can be improved by decomposing mucolytic enzyme.
