目的比较不同分期及分化程度的乙肝相关肝细胞癌中成纤维细胞激活蛋白(FAP)的表达,探讨成纤维细胞激活蛋白在肝癌中的表达差异及其意义。方法采用酶联免疫吸附方法检测成纤维细胞激活蛋白在不同分期及分化程度的乙肝相关肝细胞癌中的表达水平;用免疫组化方法对成纤维细胞激活蛋白的组织分布进行定位检查。结果成纤维细胞激活蛋白在不同分期的乙肝相关肝细胞癌组织1μg总蛋白中平均含量分别为Ⅰ期(1 039.82±248.89)pg/mL,Ⅱ期(1 173.57±184.60)pg/mL,Ⅲ期及以上(1 344.69±225.47)pg/mL,对照组为(742.15±443.16)pg/mL,差异有统计学意义(F=13.818,P=0.000);在不同分化程度的肝细胞癌组织中分别为高分化(1 008.18±148.20)pg/mL,中分化(1 126.52±218.38)pg/mL,低分化(1 414.96±311.94)pg/mL,差异有统计学意义(F=13.368,P=0.000)。免疫组化结果证实肝癌细胞中有成纤维细胞激活蛋白的表达,且其含量与肝癌细胞的病理分化程度具有明显相关性,随着肝癌细胞由高分化向低分化发展,成纤维细胞激活蛋白的表达也逐渐增加。结论成纤维细胞激活蛋白在不同分期及分化程度的乙肝相关肝细胞癌中表达存在差异,分期越晚,分化程度越低,成纤维细胞激活蛋白表达越多。
Objective To compare the expression of fibroblast activating protein (FAP) in hepatocellular carcinoma (HCC) with different stages and degree of differentiation, and to explore the difference of the expression of fibroblast activating protein in hepatocellular carcinoma and its significance. Methods The expression of fibroblast activating protein in hepatocellular carcinoma with different stages and degree of differentiation was detected by enzyme-linked immunosorbent assay (ELISA). The tissue distribution of fibroblast activating protein was detected by immunohistochemistry. Results The average content of fibroblast activating protein in 1μg total protein in different stages of hepatocellular carcinoma was stage Ⅰ (1 039.82 ± 248.89) pg / mL, stage Ⅱ (1 173.57 ± 184.60) pg / mL, stage Ⅲ (1 344.69 ± 225.47) pg / mL in the control group and (742.15 ± 443.16) pg / mL in the control group (F = 13.818, P = 0.000) The difference was statistically significant (F = 13.368, P = 0.000) for well-differentiated (1 008.18 ± 148.20) pg / mL, moderately differentiated (1 126.52 ± 218.38) pg / mL and poorly differentiated (1 414.96 ± 311.94) ). Immunohistochemistry confirmed the expression of fibroblast activating protein in hepatocellular carcinoma cells, and its content was significantly correlated with the degree of pathological differentiation of hepatoma cells. With the development of hepatocellular carcinoma cells from well-differentiated to poorly differentiated, fibroblast activating protein Expression also gradually increased. Conclusion The expression of fibroblast activating protein in hepatocellular carcinoma with different stages and degree of differentiation is different. The later stage of differentiation, the lower the degree of differentiation is, the more fibroblast activating protein is expressed.
