目的:建立高效液相色谱法测定家兔血浆中苦参碱浓度。方法:采用AICHROM-SiO2柱(250×4.6mm,5μm);流动相为甲醇:三乙胺(每100mL甲醇加3μL三乙胺);流速1.0mL/min;柱温30℃;检测波长220nm。结果:家兔血浆中苦参碱在0.1~50.0μg/mL范围内线性关系良好(r=0.9998),最低检测浓度为0.1μg/mL。低、中、高三种浓度相对回收率分别为90.0%、97.3%、97.4%,低、中、高浓度检测的日内、日间RSD分别为9.74%、6.77%、0.60%和11.5%、8.10%、0.70%。结论:该法具有操作简便可靠、准确、稳定性高等特点,可应用于血浆中苦参碱浓度的测定以及药动学研究。
Objective: To establish a method for the determination of matrine in rabbits plasma by high performance liquid chromatography. Methods: The column was AICHROM-SiO2 (250 × 4.6mm, 5μm). The mobile phase consisted of methanol: triethylamine (3μL triethylamine per 100mL of methanol), the flow rate was 1.0mL / min, the column temperature was 30 ℃, and the detection wavelength was 220nm. Results: There was a good linear relationship (r = 0.9998) between the range of 0.1 ~ 50.0 μg / mL of matrine in rabbit plasma and the lowest detection limit was 0.1 μg / mL. The relative recoveries of low, middle and high concentrations were 90.0%, 97.3% and 97.4% respectively. The intra-day and interday RSD of low, medium and high concentrations were 9.74%, 6.77%, 0.60% and 11.5%, 8.10% , 0.70%. Conclusion: The method is simple, reliable, accurate and stable. It can be applied to the determination of matrine concentration in plasma and pharmacokinetic study.
