鱼类黏液中含有许多非特异性抗菌活性成分,在自然条件下可抵抗微量病原菌的侵害,是鱼类先天性免疫系统的重要组成部分。本研究对新兴养殖鱼类星斑川鲽免疫相关组织脾脏、肾脏、后肠及皮肤黏液中抗菌物质的抗菌活性进行了研究。结果表明,星斑川鲽各免疫相关组织的提取物具有抗鳗弧菌、溶藻弧菌、副溶血弧菌和爱德华氏菌的活性。各组织的抗菌物质对温度不敏感,分别在37、50、70和95℃不同温度中处理10 min,95℃处理的样品抗菌活性显著下降,而其他温度处理后抗菌活性则变化不明显。体表黏液抗菌物质的抗菌活性在酸性条件下强于碱性条件。溶菌酶与黏液及各组织的抗菌物质作用后,其抗菌活性略有下降,胰蛋白酶和蛋白酶K处理后则没有明显的变化,并且黏液及各组织的抗菌物质具有较高的溶血活性。经凝胶过滤层析的初步鉴定结果表明,各组织的抗菌活性物质可能是分子量较小的物质。
Fish mucus contains many non-specific antimicrobial active ingredients, in natural conditions resistant to trace pathogens, is an important part of the fish’s innate immune system. In this study, the antibacterial activity of antibacterial substances in the spleen, kidney, hindgut and mucocutaneous mucosa of the newly-cultured fish Astragalus polygamis immune-related tissues was studied. The results showed that the extract of each immune-related tissue of Asteraceae salina had activity against Vibrio anguillarum, Vibrio alginolyticus, Vibrio parahaemolyticus and Edwardsiella. Antibacterial substances in each tissue were insensitive to temperature. The antibacterial activity of the samples treated with 95 ℃ for 10 min at 37, 50, 70 and 95 ℃ respectively decreased significantly, but no obvious change after the other treatments. The antibacterial activity of surface mucus antimicrobial substance is better than alkaline condition in acidic condition. The antibacterial activity of lysozyme decreased slightly after treated with mucilage and various antibacterial substances. There was no obvious change after treated with trypsin and proteinase K, and the antimicrobial substances in mucus and tissues had higher hemolytic activity. Preliminary identification by gel filtration chromatography showed that the antimicrobial actives in each tissue may be of lower molecular weight.
