目的:探讨脑康胶囊对血管性痴呆大鼠行为学及海马区凋亡基因的影响。方法:健康SD大鼠,采用双侧颈总动脉永久性结扎方法制备血管性痴呆模型,随机分为假手术组、模型组、脑康组及银杏叶提取物组。ig 4周后,对各组大鼠进行Morris水迷宫行为学测定,用酶联免疫吸附试验(ELISA)检测大鼠海马组织超氧化物歧化酶(SOD),丙二醛(MDA)表达水平;用实时荧光定量核酸扩增(q PCR),免疫印迹法(Western blot)检测B淋巴细胞瘤-2(Bcl-2),Bcl-2相关X基因(Bax)mRNA,蛋白水平及Bcl-2/Bax。结果:与假手术组比较,模型组逃避潜伏期明显延长,穿越平台次数和平台象限路径百分比明显减少;SOD活性明显降低,MDA活性明显升高;Bax mRNA与蛋白水平显著增高,Bcl-2 mRNA与蛋白水平显著降低,Bcl-2/Bax降低。与模型组比较,脑康组逃避潜伏期明显缩短,穿越平台次数和平台象限路径百分比明显增加;SOD活性明显升高,MDA含量明显降低;Bax mRNA与蛋白水平显著降低,Bcl-2 mRNA与蛋白水平显著增高,Bcl-2/Bax升高。结论:脑康胶囊可显著改善VD大鼠的学习和记忆能力,其机制可能与抗氧化应激,抑制海马区细胞凋亡有关。
Objective: To investigate the effect of Naokang Capsule on behavior and apoptosis gene in hippocampus of vascular dementia rats. Methods: The model of vascular dementia was established by permanent ligation of bilateral common carotid arteries in healthy Sprague-Dawley rats. The model was randomly divided into sham operation group, model group, Naokang group and Ginkgo biloba extract group. After 4 weeks, Morris water maze behavior was measured in each group, and the expression of superoxide dismutase (SOD) and malondialdehyde (MDA) in the hippocampus was detected by enzyme linked immunosorbent assay (ELISA); The mRNA and protein levels of Bcl-2, Bcl-2 and Bcl-2 / Bcl-2 were detected by qPCR and Western blotting. Conclusion: Nao Kang capsule can significantly improve the learning and memory ability of VD rats, and its mechanism may be related to the anti-oxidative stress and inhibition of apoptosis in hippocampus.
