目的:制备多孔羧甲基壳聚糖氟缓释膜,并对其相关性能检测和体外释药考察。方法:采用冷冻干燥法制备羧甲基壳聚糖氟缓释膜,不同比例氟化钠作为有效药物成分,并用聚乙烯吡咯烷酮作为致孔剂。以孔隙率和释氟率作为指标,筛选出最优比例;扫描电镜观察缓释膜的形态;无水乙醇液体置换法检测缓释膜的孔隙率;氟离子标准曲线考察缓释膜的体外释药性。结果:羧甲基壳聚糖/聚乙烯吡咯烷酮/氟化钠比例为1∶1∶1时,制备的多孔氟缓释膜最为理想,孔隙率为(23.4±2.3)%,释氟率为0.22 mg·d-1。结论:本实验制备的多孔羧甲基壳聚糖氟缓释膜具有孔隙率适中,释氟率稳定,体外显著延缓药物释放,并期待进一步用于覆盖义齿的根面继发龋的预防研究。
OBJECTIVE: To prepare porous carboxymethyl chitosan fluoride sustained-release membrane and investigate its related properties and in vitro release. Methods: Carboxymethyl chitosan fluoride sustained-release membrane was prepared by freeze-drying method. Different proportions of sodium fluoride were used as effective drug components and polyvinylpyrrolidone as porogen. The optimal ratio was screened by porosity and fluorine release rate, the morphology of sustained-release membrane was observed by scanning electron microscopy, the porosity of sustained-release membrane was detected by liquid-alcohol replacement method, and the standard curve of fluoride ion Medicinal properties. Results: When the ratio of carboxymethyl chitosan / polyvinylpyrrolidone / sodium fluoride was 1: 1:1, the prepared porous fluorinated sustained-release membrane was the most ideal, the porosity was (23.4 ± 2.3)% and the rate of fluorine release was 0.22 mg · d-1. CONCLUSION: The porous carboxymethyl chitosan sustained-release membrane prepared in this experiment has moderate porosity, stable rate of fluorine release and significant delay of drug release in vitro. It is also expected to be used for further prevention of secondary root caries of overdenture denture.
