目的研究青春期营养性肥胖对雄性大鼠生殖器官发育及生殖能力的影响。方法使用高脂高能量饲料喂饲4周龄Wistar雄性大鼠,制作青春期营养性肥胖模型,饲喂8周后检查大鼠生殖器官发育情况并进行病理组织学检测,采用酶联免疫吸附试验(ELISA)法检测血清睾酮、雌二醇含量,计数精子总数及活动率;按1︰2比例与雌鼠合笼1周,检测雄鼠交配指数和生育指数,并对胎鼠发育情况进行检查。结果模型组大鼠睾丸脏器系数明显低于对照组(P<0.05),光镜下发现模型组生精小管管壁变薄、细胞数减少、细胞排列紊乱;血清睾酮含量明显低于对照组(P<0.01);生育指数低于对照组(P<0.05);二组精子总数、活动率、血清雌二醇含量、交配指数、胎鼠各项发育指标比较差异均无统计学意义(P>0.05)。结论肥胖可影响大鼠睾丸发育,致血清睾酮含量降低、生育指数下降;但未观察到对胎鼠发育的影响。
Objective To study the effects of adolescent nutritional obesity on reproductive organs development and reproductive capacity in male rats. Methods Wistar male rats aged 4 weeks were fed with high-fat and high-energy diet to make adolescent nutritional obesity model. After 8 weeks of feeding, the reproductive organs of rats were examined for pathological and histological examination. Enzyme-linked immunosorbent assay ELISA method was used to detect serum testosterone and estradiol levels and count the total number of sperm and activity rate. According to the ratio of 1: 2, the mice were caged for 1 week. The mate index and fertility index of males were detected. Results The testicular organ coefficient of model group was significantly lower than that of the control group (P <0.05). Under the light microscope, the wall of the seminiferous tubule in the model group was thinner, the cell number decreased and the cell arrangement was disorganized. The serum testosterone content was significantly lower than that of the control group (P <0.01). The fertility index was lower than that of the control group (P <0.05). There was no significant difference in sperm count, activity rate, serum estradiol level, mating index and fetal development between the two groups > 0.05). Conclusion Obese can affect the development of testis in rats, resulting in the decrease of serum testosterone and the decrease of fertility index. However, no effect on fetal rat development was observed.
