目的观察丹红注射液对实验性脑出血大鼠神经功能评分和血清TNF-α、CRP、IL-6水平的影响,探讨其保护神经功能的可能机制。方法 30只健康Wistar雄性大鼠随机分为假手术组、模型组和丹红组。运用脑内自身动脉血注入法建立脑出血大鼠模型,假手术组只进针不注血,丹红组造模前后予腹腔注射丹红注射液(2 ml·kg~(-1)·d~(-1)),假手术组和模型组予腹腔注射生理盐水2 ml·kg~(-1)·d~(-1),均1次/24 h。分别于实验1、3、5和7 d以ELISA法测定血清肿瘤坏死因子(TNF)-α、白介素(IL)-6)、C-反应蛋白(CRP)的水平。按Berderson标准,于术前和术后行神经功能缺损评分。结果模型组和丹红组神经功能缺损评分及TNF-α、CRP、IL-6水平明显高于假手术组(P均<0.05),丹红组神经功能缺损评分及TNF-α、CRP、IL-6水平明显低于模型组(P均<0.05)。结论丹红注射液有保护脑出血大鼠神经功能的作用,可能与降低TNF-α、IL-6、CRP在血清中的释放有关。
Objective To observe the effect of Danhong injection on neurological function score, serum TNF-α, CRP and IL-6 levels in rats with experimental intracerebral hemorrhage (ICH) and to explore the possible mechanism of Danhong injection in protecting neurological function. Methods Thirty healthy Wistar male rats were randomly divided into sham operation group, model group and Danhong group. The model of intracerebral hemorrhage was established by injecting intracerebral arterial blood into the rats. The rats in sham operation group were injected with Danhong injection (2 ml · kg ~ (-1) d ~ (-1)). The rats in sham operation group and model group were intraperitoneally injected with normal saline (2 ml · kg ~ (-1) · d ~ (-1)) once every 24 hours. Serum levels of tumor necrosis factor (TNF) -α, interleukin (IL) -6) and C-reactive protein (CRP) were measured by ELISA at 1, 3, 5 and 7 days. According to Berderson criteria, neurological deficit scores were performed preoperatively and postoperatively. Results The neurological deficit score and the levels of TNF-α, CRP and IL-6 in the model group and the Danhong group were significantly higher than those in the sham operation group (all P <0.05). The neurological deficit score and TNF-α, CRP, IL -6 levels were significantly lower than the model group (P <0.05). Conclusion Danhong injection can protect the neurological function of rats with intracerebral hemorrhage, which may be related to the decrease of the release of TNF-α, IL-6 and CRP in serum.
