浙江省近几年甲_3型流行性感冒病毒株HA_1基因特性分析

为了研究近几年浙江省甲3 型流行性感冒 (流感 )流行株血凝素 (HA)基因的特性 ,阐明HA基因的变异与流感流行的关系。对浙江省 1998~ 2 0 0 2年流感流行期间分离的甲3 型代表性毒株 ,提取病毒核糖核酸 (RNA) ,采用逆转录 聚合酶链反应 (RT PCR)扩增病毒HA1基因 ,纯化产物进行核苷酸序列测定 ,并用DNAMAN和BioEdit软件作分析处理。结果显示 :浙江省近几年流感流行期间分离到的甲3 型流感病毒中的 8株代表毒株 ,其在HA1区域的核苷酸长度均为 987bp ,由此推导的氨基酸数为 32 9个。A/浙江 / 2 5 / 98与当时世界卫生组织 (WHO)推荐的流感疫苗株A/武汉 / 35 9/ 95株在HA1区域的核苷酸同源性为 97 4 % ,氨基酸同源性仅为 94 8% ;HA1区的抗原性已发生了一定的变异。A/浙江 / 11/ 0 2株与同期WHO推荐的流感疫苗株A/莫斯科 / 10 / 99相比 ,两者的氨基酸同源性仅为 96 1% ;且氨基酸的改变发生在抗原决定簇A区和B区的有 4个位点 ,与 1998年的流行株A/浙江 / 2 5 / 98的氨基酸同源性也只有 96 1%。说明A/浙江 / 11/ 0 2株与A/莫斯科 / 10 / 99和A/浙江 / 2 5 / 98株相比 ,其HA1区的抗原性也发生了变化 ,在基因进化树上远离A/莫斯科 / 10 / 99株和A/浙江 / 2 5 / 98株。表明由于甲3 型流感病毒HA1区域的抗原漂移导 In order to study the characteristics of hemagglutinin (HA) gene of epidemic strains of influenza A (influenza) in Zhejiang Province in recent years, the relationship between HA gene variation and influenza prevalence was elucidated. The virus RNA (RNA) was extracted from the representative strain of type A 3 isolated during the influenza epidemic in Zhejiang province from 1998 to 2002. The virus HA1 gene was amplified by reverse transcription polymerase chain reaction (RT PCR) Nucleotide sequencing was performed and analyzed using DNAMAN and BioEdit software. The results showed that 8 representative strains of influenza A virus isolated from influenza in recent years in Zhejiang Province were all 987 bp in HA1 region and deduced 32 9 amino acids . A / Zhejiang / 2 5/98 and the then WHO vaccine strain A / Wuhan / 359/95 that were recommended by WHO at that time had 97 4% nucleotide homology in the HA1 region and only 94% homology in amino acid homology 94 8%. The antigenicity of HA1 region had some variation. A / Zhejiang / 11/0 2 strains compared with the WHO recommended influenza vaccine strain A / Moscow / 10/99 in the same period, the amino acid homology between the two strains was only 96.1%; and amino acid changes occurred in the antigenic determinant A There were 4 loci in area B and area B, and only 96 1% amino acid homology with the 1998 epidemic strain A / Zhejiang / 25/98. The results showed that the antigenicity of HA1 in A / Zhe Jiang / 11/0 changed compared with that in A / Moscow / 10/99 and A / Zhejiang / 25/98, Moscow / 10/99 and A / Zhejiang / 2 5/98 strains.
