目的研究α-细辛醚对癫痫大鼠的海马钙稳态的影响。方法大鼠癫痫持续状态(SE)后,观察α-细辛醚对海马游离Ca2+浓度、游离钙调蛋白(CaM)平均通道荧光值、总CaM及钙调蛋白依赖性蛋白激酶(CaMK)IIa-mRNA表达的变化的影响。结果模型组SE后,各时相海马游离Ca2+显著增高(P<0.05);CaM平均通道荧光值显著降低(P<0.05);SE后12~72 h,游离CaM表达较正常组明显增强(P<0.05);CaMK IIa-mRNA表达显著降低(P<0.05)。与模型组相比,α-细辛醚组游离Ca2+浓度显著降低,游离CaM平均通道荧光值明显增加,总CaM表达显著降低,CaMK IIa-mRNA表达明显增强(P<0.05)。结论α-细辛醚可下调SE后海马游离Ca2+浓度及结合CaM表达,并上调CaMK IIa-mRNA表达。
Objective To study the effects of α-asarone on hippocampal calcium homeostasis in epileptic rats. Methods After the status epilepticus (SE) in rats, the effects of α-asarone on the free Ca 2+ concentration, the average fluorescence intensity of free calmodulin (CaM), total CaM and CaMK IIa- Effect of changes in mRNA expression. Results After SE, the levels of free Ca2 + in hippocampus were significantly increased at each time point (P <0.05), and the fluorescence intensity of CaM was significantly decreased (P <0.05). After 12-72 h of SE, the expression of free CaM was significantly increased <0.05). The expression of CaMK IIa-mRNA was significantly decreased (P <0.05). Compared with the model group, the concentration of free Ca2 + in α-asarone group decreased significantly, the mean fluorescence of free CaM increased significantly, the total CaM decreased significantly, and the expression of CaMK IIa-mRNA increased significantly (P <0.05). Conclusion α-asarone can down-regulate SE after hippocampal free Ca2 + concentration and binding CaM expression, and up-regulate CaMK IIa-mRNA expression.
