目的探讨单采血小板(A-PLT)和手工分离血小板(BC-PLT)在保存过程中,去血小板血浆中可溶性CD40配体(sCD40L)含量变化和区别。探不讨同保存时间血小板的去血小板血浆及其中的sCD40L对中性粒细胞(PMN)呼吸爆发影响。方法取5份同1人份不同保存时间1d、3d和5d单采血小板和手工分离血小板,离心分离得到去血小板血浆。ELISA检测去血小板血浆中sCD40L的含量。二氢若丹明123(DHR)法流式检测保存1d、3d和5d去血小板血浆对PMN呼吸爆发的影响。通过抗sCD40L抗体抑制血浆中sCD40L和过滤去除血浆中的血小板微粒,流式检测sCD40L对PMN呼吸爆发的影响。
Objective To investigate the changes and differences of soluble CD40 ligand (sCD40L) in platelet-depolymerized plasma (PLT) and manual isolated platelets (BC-PLT) during preservation. Probe into the effect of sCD40L on platelet depolarization and neutrophil (PMN) respiratory burst. Methods Five copies of apheresis platelets and manual detached platelets were collected on day 1, day 3, and day 5 at the same retention time of 1 human. Platelets were separated by centrifugation. ELISA assay of platelet-derived plasma sCD40L levels. Effects of Dehydrated Rhodamine 123 (DHR) Flow Assay on PMD Respiratory Burst in 1d, 3d and 5d Depolarized Platelets. Anti-sCD40L antibody was used to inhibit sCD40L in plasma and filtration to remove platelet particles in plasma, and flow cytometry was used to detect the effect of sCD40L on PMN respiratory burst.
