目的阐述端粒酶的结构、功能以及端粒酶与肝纤维化的关系,探讨肝纤维化过程中肝细胞与肝星状细胞内端粒酶活性的变化,为肝纤维化的防治提供新的药物作用靶点。方法依据近年国内外的相关文献对端粒酶相关系统进行综述。结果在肝纤维化形成过程中,肝细胞中端粒是缩短的,从端粒酶的经典与非经典的生理功能角度上增加端粒酶的活性可使肝细胞获得再生的能力;肝星状细胞中的端粒是延长的,从端粒酶的经典生理功能角度上减弱端粒酶的活性可使肝星状细胞凋亡。结论端粒酶有望成为治疗肝纤维化的一个潜在的药物靶点。
OBJECTIVE: To elucidate the relationship between telomerase activity and hepatic fibrosis in order to explore the changes of telomerase activity in hepatocytes and hepatic stellate cells during liver fibrosis, and to provide a new method for the prevention and treatment of hepatic fibrosis Drug target. Methods Based on the related literatures at home and abroad in recent years, the related systems of telomerase were reviewed. Results During hepatic fibrosis, telomeres were shortened in hepatocytes. Increasing the activity of telomerase from the classical and non-classical physiological functions of telomerase resulted in the ability of hepatocytes to regenerate. Telomeres in cells are prolonged, and telomerase can attenuate hepatic stellate cells by attenuating telomerase activity from the classical physiological functions of telomerase. Conclusion Telomerase is expected to be a potential drug target for the treatment of hepatic fibrosis.
