目的:探讨p38MARK蛋白表达在槲皮素对镉致急性肾损伤保护机制中的作用。方法:雄性Wismr大鼠随机分成对照组、镉组、治疗组(再根据槲皮素剂量分为高、中、低剂量组)。对大鼠分别给予生理盐水+盐水、氯化镉+盐水、氯化镉+槲皮素,30天后处死大鼠,取肾脏组织。对所取肾脏组织进行western blot检测,观察肾脏p38MAPK的蛋白表达。结果:镉组与对照组相比p38MAPK蛋白表达明显增加(p<0.01):治疗组低、中、高3组与镉组相比p38MAPK蛋白的表达均降低(p<0.05,p<0.01),且3组之间相比随着槲皮素剂量的增加p38MAPK蛋白表达具有下降趋势(p<0.05,P<0.01)。结论:p38MARK参与了槲皮素对镉致急性肾损伤的保护作用;槲皮素可以抑制镉致急性肾损伤过程中p38MARK蛋白的表达:槲皮素是通过p38MARK信号途径而发挥其对镉致急性肾损伤的保护作用。
AIM: To investigate the role of p38MARK protein in the protective mechanism of quercetin on acute kidney injury induced by cadmium. Methods: Male Wismr rats were randomly divided into control group, cadmium group and treatment group (divided into high, medium and low dose groups according to quercetin dose). Rats were given saline + saline, cadmium chloride + saline, cadmium chloride + quercetin, 30 days after the rats were sacrificed, take the kidneys. The kidneys were taken for western blot detection and the protein expression of p38MAPK in kidney was observed. Results: Compared with the control group, p38MAPK protein expression was significantly increased in the cadmium group (p <0.01). The expression of p38MAPK protein in the low, middle and high groups was lower than that in the cadmium group (p <0.05, p <0.01) Compared with the control group, p38MAPK protein expression decreased with quercetin dosage increasing (p <0.05, P <0.01). CONCLUSIONS: p38MARK is involved in the protective effect of quercetin on acute kidney injury induced by cadmium. Quercetin can inhibit the expression of p38MARK protein in acute kidney injury induced by cadmium. Quercetin exerts its acute toxicity to cadmium through p38MARK signal pathway Kidney injury protection.
