目的:研究原青花素对特非那定诱导斑马鱼的心脏损伤的保护作用,为原花青素的心脏保护作用的开发提供依据。方法:选取发育正常的48hpf(hour post fertilization)AB系斑马鱼胚胎,以0.5、1、3和5mg/L的原花青素对用特非那定(0.94mg/L)诱导斑马鱼心脏形态和功能影响的考察,在72hpf时,观察斑马鱼的心脏相关的指标,包括心脏形状、心囊大小、血液循环、血细胞在心区堆积等情况,测量斑马鱼的心率、测量静脉窦-动脉球(SV-BA)间距,以及主动脉的血流速度。结果:特非那定处理斑马鱼后,斑马鱼心脏出现了心脏形态、心率、功能等改变。原花青素达到1mg/L时可明显改善心脏泵血和全身血流速度等功能方面的作用;原花青素增加到3 mg/L时可明显缩短斑马鱼的SV-BA间距,缓解血细胞在心区堆积、心囊水肿等形态学的变化;原花青素增加至5mg/L时可明显提升斑马鱼心率。结论:原花青素对斑马鱼心脏功能损伤具有一定的保护作用,本实验为原花青素在心保护性作用药物的开发提供依据。
