目的 :观察人工合成的 β淀粉样多肽 (Aβ)对培养神经元细胞内游离钙 ([Ca2 + ]i)的影响以及褪黑素 (Mel)的阻断作用。材料和方法 :用激光扫描共聚焦显微镜技术 ,检测给予不同浓度 (Aβ2 5~ 3 5和Mel后新生大鼠皮层培养神经元 [Ca2 + ]i荧光强度。结果 :神经元暴露Aβ2 5~ 3 50 5、 1、 10 μmol/L环境后 ,[Ca2 + ]i荧光与对照组比较均呈较有意义升高 (P <0 0 5 ) ;同时暴露Mel组 ,未见钙荧光强度升高。结论 :Aβ2 5~ 3 5可诱导神经元[Ca2 + ]i升高 ,并呈剂量依赖性 ;预先给予Mel可部分阻断这种毒性作用。
Objective: To observe the effects of synthetic β-amyloid polypeptide (Aβ) on intracellular free calcium ([Ca2 +] i) and the blocking effect of melatonin (Mel) in cultured neurons. MATERIALS AND METHODS: The fluorescence intensity of [Ca2 +] i in cortical neurons exposed to Aβ2 5 ~3 5 and Mel was detected by laser scanning confocal microscopy.Results: Neuronal exposure to Aβ2 5 ~ 3 50 Compared with the control group, the fluorescence of [Ca2 +] i increased more significantly (P <0.05) at 5, 1 and 10 μmol / L, and no increase of calcium fluorescence was observed in the Mel group. : Aβ2 5 ~ 3 5 can induce neuronal [Ca2 +] i increased in a dose-dependent manner; pre-given Mel can be partially blocked this toxic effect.
