目的:研究巨细胞病毒(CMV)宫内感染对子代豚鼠海马神经元Tau蛋白磷酸化的影响。方法:建立豚鼠CMV宫内感染动物模型,PCR法筛选CMV DNA阴性豚鼠,雌雄同笼受孕;孕龄1-20d时,随机分成两组:感染组腹腔内接种CMV病毒悬液,对照组接种生理盐水,足月分娩的子代豚鼠喂养至3月龄后无菌采集脑组织,应用免疫组织化学法和免疫印迹法检测各组子代豚鼠海马神经元Tau、p-Tau(Ser 404)、PKA及PP2A蛋白表达水平。结果:与对照组相比,感染组子代豚鼠海马神经元Tau、p-Tau(Ser 404)及PKA蛋白的表达水平明显上升,而PP2A蛋白的表达水平明显下降,差异具有统计学意义(P<0.01)。结论:CMV宫内感染导致子代豚鼠海马神经元Tau蛋白磷酸化水平增加,可能与PKA及PP2A蛋白的表达异常相关。
Objective: To study the effect of cytomegalovirus (CMV) intrauterine infection on Tau phosphorylation in hippocampal neurons of offspring guinea pigs. Methods: CMV intrauterine infection in animal models was established. CMV DNA-negative guinea pigs were screened by PCR. The pregnant women were males and females at the same time. The pregnant women were randomly divided into two groups at 1 to 20 days. CMV virus suspension was inoculated intraperitoneally in control group, Brine, term offspring of guinea pigs were fed to 3 months after aseptic collection of brain tissue, immunohistochemistry and Western blot were used to detect the hippocampal neurons of each group offspring Tau, p-Tau (Ser 404), PKA And PP2A protein expression levels. Results: Compared with the control group, the expression levels of Tau, p-Tau (Ser 404) and PKA protein in hippocampal neurons of guinea pigs were significantly increased, while the expression of PP2A protein was significantly decreased (P <0.01). CONCLUSION: Intrauterine infection of CMV leads to an increase of Tau phosphorylation in hippocampal neurons of offspring guinea pigs, which may be related to the abnormal expression of PKA and PP2A protein.
