前单核细胞 THP- 1分化是一个复杂的过程 ,这一过程包含了复杂的细胞内信号传导及许多因子的活化及大量蛋白的表达。基质金属蛋白酶 - 9(Motrix Metalloproteinase- 9,MMP- 9)主要降解基底膜中的 和 型胶原 ,在血管增生及肿瘤浸润转移过程中起重要的作用。为探讨核转录因子 κB(nuclear factor kappaB,NF- κB)在 THP- 1粘附及 MMP- 9表达中的作用 ,我们应用 NF- κB抑制剂 PDTC(Pyrollidine Dithiocarba-mate二硫碳吡咯烷 ,应用共聚焦显微镜、流式细胞术观测了其对 THP- 1粘附及 MMP- 9表达的影响。结果表现 :PDTC可明显的抑制 THP- 1的粘附及 MMP- 9的表达。表明 NF- κB在细胞粘附及 MMP- 9的表达中起重要的作用。
Differentiation of pre-monocytes THP-1 is a complex process that involves complex intracellular signaling and the activation of many factors and the expression of large amounts of protein. Matrix metalloproteinase-9 (MMP-9) mainly degrades collagen in the basement membrane and plays an important role in angiogenesis and tumor invasion and metastasis. To investigate the role of nuclear factor kappaB (NF-κB) in the adhesion of THP-1 and the expression of MMP-9, we investigated the effects of NF-κB inhibitor PDTC (Pyrollidine Dithiocarba-mate) Confocal microscopy and flow cytometry were used to observe the effect of THP-1 on the adhesion and the expression of MMP-9 .Results: PDTC could significantly inhibit the adhesion of THP-1 and the expression of MMP-9, indicating that NF- Plays an important role in cell adhesion and MMP-9 expression.
