目的研究哺乳期BDE-153暴露对大鼠成年后大脑皮质胆碱能系统关键酶的活力及细胞内Ca2+浓度的影响。方法将40只出生第4天的雄性清洁级SD大鼠按体重随机分成4组,分别为溶剂对照(橄榄油)组和低(1 mg/kg)、中(5 mg/kg)、高剂量(10 mg/kg)BDE-153染毒组,每组10只。在仔鼠出生第10天,采用一次性腹腔注射方式进行染毒,染毒容量为0.01 ml/g。染毒两个月后,采用流式细胞仪测定大脑皮质细胞内Ca2+的浓度,采用比色法测定皮质乙酰胆碱转移酶(ChAT)和乙酰胆碱酯酶(TchE)的活力。结果与溶剂对照组比较,各剂量BDE-153染毒组大鼠大脑皮质神经细胞内Ca2+浓度均降低,差异有统计学意义(P<0.05);仅高剂量BDE-153染毒组皮质组织内ChAT和TchE活力降低,差异均有统计学意义(P<0.05)。结论哺乳期BDE-153暴露可抑制成年大鼠大脑皮质胆碱能系统的功能,使细胞内Ca2+浓度降低,这可能是BDE-153神经毒性的主要机制之一。
Objective To investigate the effect of lactation on BDE-153 exposure on the key enzyme activity and intracellular Ca2 + concentration in the adult rat cholinergic system. Methods Forty male SD rats aged 4 days were randomly divided into four groups according to body weight: solvent control (olive oil) group and low (1 mg / kg), medium (5 mg / kg) (10 mg / kg) BDE-153 group, 10 rats in each group. The pups were born on the 10th day, using a one-time intraperitoneal injection of exposure, exposure capacity of 0.01 ml / g. Two months after exposure, the concentration of Ca2 + in cerebral cortical cells was determined by flow cytometry. The viability of cortical acetylcholinesterase (ChAT) and acetylcholinesterase (TchE) was measured by colorimetry. Results Compared with the solvent control group, the concentration of Ca2 + in cerebral cortical neurons of BDE-153 group decreased significantly at each dose (P <0.05), and only in the high dose BDE-153 group ChAT and TchE activity decreased, the differences were statistically significant (P <0.05). Conclusion BDE-153 exposure during lactation can inhibit the function of the cerebral cortex cholinergic system in adult rats and decrease the intracellular Ca2 + concentration, which may be one of the main mechanisms of BDE-153 neurotoxicity.
