目的:探讨凋亡在视神经撞击和挤压损伤导致视网膜神经节细胞死亡中的作用。方法:应用活细胞悬液荧光染色法和流式细胞仪 P I染色法检测损伤后视网膜神经元凋亡的形态和数量。结果:荧光染色法见视野内有散在细胞出现核染色质凝聚现象( 不规则核块) ,流式细胞仪 D N A 直方图见正常对照组有直而高的 G0/ G1 峰和低平的 G2 峰,在 G0/ G1 之前有一细胞群体,大约占细胞总数的163 % ,损伤组在 G0/ G1 之前有一低小的亚二倍体峰,大约占全部细胞的483 % 。结论:视神经不完全损伤后存在视网膜神经元的凋亡,这一结论可为视神经外伤的临床治疗提供实验依据。
Objective: To investigate the role of apoptosis in retinal ganglion cell death caused by optic nerve crush and crush injury. Methods: The morphology and quantity of apoptotic retinal neurons apoptosis were detected by fluorescence staining of live cell suspension and flow cytometry P I staining. Results: Focal chromatin condensation phenomenon (irregular nuclei) appeared in the scattered cells in the field of vision. The histone D N A histogram of the flow cytometry showed a straight and high G0 / G1 peak and a low level G2 peak, there is a cell population before G0 / G1, accounting for about 1.63% of the total number of cells. The injury group has a small sub-diploid peak before G0 / G1, accounting for about 4.83% . Conclusion: There is apoptosis of retinal neurons after incomplete optic nerve injury. This conclusion may provide experimental evidence for the clinical treatment of optic nerve trauma.
