目的:研究山芝麻水提物(helicteres angustifolia)体内抗鸭乙型肝炎病毒(duck hepatitis B virus,DHBV)的作用。方法:将DHBV-DNA阳性麻鸭随机分为山芝麻高、中、低剂量组(10,5,2.5 g.kg-1)、拉米夫定50 mg.kg-1组和模型组,分别给予相应药物进行干预。用药前、用药第7,14天及停药第7天,取静脉血用实时荧光定量PCR法检测血清DHBV-DNA含量。治疗后取肝脏组织作病理学检查。结果:拉米夫定组用药后血清DHBV-DNA水平迅速降低,停药后立即反跳;山芝麻高、中剂量组用药7,14 d血清中的DHBV-DNA显著降低(P<0.05或P<0.01),停药后仍表现有持续的抑制作用;在给药及停药过程中山芝麻低剂量组血清DHBV-DNA变化不明显。镜下观察显示山芝麻高剂量组与阳性药组鸭肝脏的病理改变有一定的改善。结论:山芝麻在鸭体内有一定的抑制鸭乙型肝炎病毒DNA的作用,其作用有明显的量效和时效反应关系。
Objective: To study the anti-duck hepatitis B virus (DHBV) effect in helicteres angustifolia. Methods: DHBV-DNA positive ducks were randomly divided into high, medium and low doses of sesame (10,5,2.5 g.kg-1), lamivudine 50 mg.kg-1 and model group Give the appropriate drug to intervene. Before treatment, on the 7th and 14th day of treatment and on the 7th day of withdrawal, the content of DHBV-DNA in serum was detected by real-time fluorescence quantitative PCR. Liver tissue after treatment for pathological examination. Results: Serum DHBV-DNA levels in the lamivudine group decreased rapidly and rebounded immediately after stopping the treatment. DHBV-DNA in the sera of the high and middle-dose sesame groups was significantly lower (P <0.05 or P <0.01). After discontinuation, there was still a continuous inhibition. Serum DHBV-DNA in the low-dose sesame seeds group was not significantly changed during administration and withdrawal. Microscopic observation showed that the high-dose sesame mountain group and the positive group of duck liver pathological changes have some improvement. Conclusion: Mountain sesame can inhibit the DNA of duck hepatitis B virus in duck body, and its effect has obvious dose-effect and time-effect relationship.
