目的研究灯盏乙素对过氧化氢(H2O2)诱导PC12细胞损伤的保护作用。方法建立H2O2致PC12细胞损伤模型,倒置相差显微镜下进行一般形态学观察,化学比色法测定乳酸脱氢酶(LDH)释放量及细胞培养液和细胞内丙二醛(MDA)含量和超氧化物歧化酶(SOD)活性。结果100μmol/L H2O2诱导PC12细胞4 h,细胞呈现明显损伤形态,LDH释放量增加,细胞培养液和细胞内MDA含量升高,SOD活性降低。灯盏乙素可明显改善形态学损伤,显著降低LDH释放量、细胞培养液及细胞内MDA含量,提高SOD活性。结论灯盏乙素对H2O2诱导PC12细胞损伤具有保护作用,其作用机制可能与提高PC12细胞的抗氧化能力有关。
Objective To investigate the protective effect of scutellarin on hydrogen peroxide (H2O2)-induced injury in PC12 cells. Methods The injury model of PC12 cells induced by H2O2 was established. The morphology was observed under an inverted phase contrast microscope. The amount of lactate dehydrogenase (LDH) released, the content of malondialdehyde (MDA) in the cell culture medium and the cells, and the hyperoxidation were determined by chemical colorimetry. Dismutase (SOD) activity. Results After 100 μmol/L H2O2 induced PC12 cells for 4 h, the cells showed obvious damage morphology, LDH release increased, cell culture medium and intracellular MDA content increased, SOD activity decreased. Scutellarin can significantly improve morphological damage, significantly reduce the release of LDH, cell culture fluid and intracellular MDA content, and increase SOD activity. Conclusion The scutellarin can protect PC12 cells from injury induced by H2O2, and its mechanism may be related to the increase of the antioxidant capacity of PC12 cells.
