传统藏药“解吉”主要基原植物之一粗壮秦艽Gentiana robusta分布区广,且与近缘种呈同域分布现象,遗传背景复杂。多居群平行取样,直接测定该物种及其近缘物种麻花艽G.straminea、粗茎秦艽G.crassicaulis、长梗秦艽G.waltonii,外类群椭圆叶花锚Halenia elliptica的r DNA ITS区序列及叶绿体DNA的mat K,rbc L,rpo C1,trn L(UAA),psb A-trn H,atp B-rbc L,trn S(GCU)-trn G(UCC),rpl20-rps12,trn L(UAA)-trn F(GAA)序列228条;TA克隆法测定粗壮秦艽ITS区序列120条。对粗壮秦艽进行基因分型,形态学及DNA分子证据显示其可能为一杂交物种;克隆序列中寻找稳定位点,多片段组合可鉴定粗壮秦艽及其近缘种。该文对遗传背景多样的药用植物鉴定研究具有重要参考价值。
Gentiana robusta, one of the major primitive plants of traditional Tibetan medicine “Jieji”, is widely distributed and has the same-domain distribution with related species, with a complex genetic background. The multi-population parallel sampling was used to directly determine the rDNA ITS sequence of this species and its related species, G. straminea, G. crassicaulis, G. waltonii, Halenia elliptica and chloroplast DNA Matk rbc L rpo C1 trn L (UAA) psb A-trn H atp B-rbc L trn S (GCU) -trn G (UCC) rpl20-rps12 trn L (UAA) - 228 sequences of trn F (GAA) were sequenced. TA cloning was used to determine 120 ITS sequences in Gentiana macrophylla. Genotyping of stout Gentiana macrophylla, morphological and molecular evidence of DNA showed that it may be a hybrid species; looking for stable loci in the cloned sequences, multi-fragment combinations can identify stout Gentiana macrophylla and its related species. This article has important reference value for the identification of medicinal plants with diverse genetic backgrounds.
