目的:探讨矢车菊素-3-葡萄糖苷对体内外卵巢癌生长的影响及其机制。方法:矢车菊素-3-葡萄糖苷作用人卵巢癌细胞株HO-8910PM后,CCK-8法检测细胞增殖;流式细胞术检测细胞凋亡;Western blotting检测卵巢癌细胞中Mucin-4蛋白表达;建立起裸鼠卵巢癌皮下移植瘤模型,观察矢车菊素-3-葡萄糖苷对裸鼠卵巢癌皮下移植瘤生长的影响;免疫组织化学法检测肿瘤组织中Ki-67和Mucin-4的阳性表达。结果:与对照组相比较,矢车菊素-3-葡萄糖苷可显著抑制体外卵巢癌HO-8910PM细胞生长,并明显诱导细胞凋亡;可显著下调卵巢癌细胞中Mucin-4的表达。矢车菊素-3-葡萄糖苷可显著抑制裸鼠卵巢癌皮下移植瘤生长;可明显降低卵巢癌肿瘤组织中Ki-67和Mucin-4的阳性表达。结论:矢车菊素-3-葡萄糖苷可显著抑制体内外卵巢癌生长,该作用可能通过抑制Mucin-4蛋白表达而实现。
Objective: To investigate the effects of cyanidin-3-glucoside on the growth of ovarian carcinoma in vitro and in vivo and its mechanism. Methods: The proliferation of human ovarian cancer cell line HO-8910PM was induced by cyanidin-3-glucoside, the cell proliferation was detected by CCK-8 assay, the apoptosis was detected by flow cytometry, the expression of Mucin-4 protein by Western blotting The effect of cyanidin-3-glucoside on the growth of subcutaneously transplanted ovarian cancer in nude mice was observed. The expressions of Ki-67 and Mucin-4 in tumor tissues were detected by immunohistochemistry Positive expression. Results: Compared with the control group, cyanidin-3-glucoside significantly inhibited the growth of ovarian cancer HO-8910PM cells and induced the apoptosis of ovarian cancer HO-8910PM cells. The expression of Mucin-4 in ovarian cancer cells was significantly decreased. Cyanidin-3-glucoside can significantly inhibit the growth of subcutaneously transplanted ovarian cancer in nude mice; It can significantly reduce the positive expression of Ki-67 and Mucin-4 in ovarian cancer. CONCLUSION: Cyanidin-3-glucoside can significantly inhibit the growth of ovarian cancer in vitro and in vivo, and this effect may be achieved by inhibiting the expression of Mucin-4 protein.
