目的:分析生殖细胞特异基因-2(GSG2)在肺癌组织的预后意义,探讨其mRNA的表达水平与患者常见肺癌临床病理学特点的相关性。方法:在线生存信息分析网站中研究肺癌组织中的GSG2的mRNA水平对肺癌患者生存的影响。收集苏州大学附属第一医院2018年1月到2019年2月间胸外科的15例肺癌患者手术切除后的新鲜冰冻标本,应用实时定量聚合酶链反应(Real-time PCR)定量分析肺癌组织和正常肺组织的GSG2的表达;扩大样本量后,在86例肺癌患者中,分析GSG2的表达水平和肺癌临床病理学特点的关系。肺癌癌组织和正常组织的GSG2表达差异采用配对n t检验,GSG2的mRNA表达量和肺癌常见临床病理学特征的相关性采用Fisher’s确切概率法或者检验分析。n 结果:GSG2高表达的肺癌人群的总生存期(OS)在总体人群、腺癌患者、女性患者、男性患者和吸烟者中明显低于GSG2低表达者(n P<0.05);GSG2高表达的肺癌人群的首次进展生存(FP)在总体人群、男性和吸烟者中显著低于GSG2低表达组(n P<0.05);在肺癌新鲜冰冻组织中的GSG2的表达水平显著高于正常肺组织(2.15±0.34比1.00±0.12,n P<0.05);肺癌中GSG2的表达水平与肺癌的淋巴结分期呈明显正相关(n P<0.05),较高的GSG2表达水平提示较多的淋巴结转移。n 结论:较高的GSG2提示肺癌患者较差的预后,同时GSG2在肺癌组中显著高表达,与肺癌淋巴结转移明显相关。“,”Objective:To test the expression of germ cell specific gene 2 (GSG2) mRNA in lung cancer tissue samples, and analyze the effects of GSG2 on survival of lung cancer patients and the correlation between GSG2 and the clinicopathological characteristics in lung cancer.Methods:Online Kaplan-Meier database was used to evaluate the effects of GSG2 on lung cancer overall survival (OS) and first progression (FP). Real-time quantitative polymerase chain reaction (Real-time PCR) was utilized to evaluate the expression levels of GSG2 in cancer and normal adjacent tissues in lung cancer. The relationship between GSG2 mRNA and the clinical and pathological features in lung cancer was analyzed.Results:OS rate of lung cancer patients with higher levels of GSG2 were shorter than those with lower levels of GSG2 expression in all patients, adenocarcinoma, female patients, male patients and smoking groups (n P<0.05). Similar trends were also observed in FP analysis in all patients, male patients and smoking groups (n P<0.05). The GSG2 levels were higher in lung cancer tissues than those in normal tissues (2.15±0.34 vs. 1.00±0.12,n P<0.05). GSG2 levels were correlated to N stage positively (n P<0.05).n Conclusion:Increased expression of GSG2 is associated with shorter survival in lung cancer. GSG2 was higher in lung cancer tissues, and related to lymph node metastasis positively.
