本研究以单株马尾松的142个大配子体为作图群体,利用筛选的55对SSR引物和142对SRAP引物获得502个标记,构建了一张包含237个标记、18个连锁群的马尾松遗传连锁图谱。马尾松连锁图谱总图距为1 462.1 cM,最大连锁群图距为136.4 cM,最小连锁群图距为36.7 cM,平均每个连锁群长度为81.2 cM,标记之间平均图距为6.2 cM。此外,利用L16(45)正交试验表对影响PCR结果的Mg2+、dNTP、模板DNA、引物浓度、Taq DNA聚合酶进行试验,建立了适合马尾松SSR-PCR和SRAP-PCR的反应体系。
In this study, 142 large gametophytes of Pinus massoniana were used as the mapping population. Fifty-two SSR primers and 142 pairs of SRAP primers were screened for 502 markers, and a 237-marker massoniana massoniana Genetic linkage map. The total length of linkage map of Masson pine was 1 462.1 cM, the maximum linkage map distance was 136.4 cM, the minimum linkage map distance was 36.7 cM, the average length of each linkage group was 81.2 cM, and the average distance between markers was 6.2 cM. In addition, the reaction system of SSR-PCR and SRAP-PCR for Masson pine was established by using L16 (45) orthogonal test table to test Mg2 +, dNTP, template DNA, primer concentration and Taq DNA polymerase.
