Temperature-controlled high performance liquid chromatography is a new means of exploring human genomic DNA sequence variation. Temperature-controlled high-performance liquid chromatography can detect DNA variability sensitively and accurately by comparing heteroduplexes to homoduplexes rather than sequencing samples one-by-one, with the only identified mutations requiring further sequencing. This approach can significantly reduce the sequencing effort for topics that seek to discover new mutations or new polymorphisms, while significantly reducing the costs associated with detecting fewer variants. By automating operations, the speed at which data can be accessed can be dramatically increased. This article describes the principles and nomenclature of temperature-controlled high performance liquid chromatography, its major advantages and applications
