以人弥漫大B细胞淋巴瘤(diffuse large B-cell lymphoma,DLBCL)活化B细胞(activated B-cell,ABC)型细胞株OCI-Ly3为研究对象,探索ABC型的生物学特性及肿瘤相关分子的表达,以及小鼠成瘤条件.对不同条件下培养的OCI-Ly3细胞进行对比分析,观察细胞生长形态及生物学特性;分析肿瘤相关抗原在OCI-Ly3细胞的表达情况;采用皮下和腹腔接种,在SCID小鼠中建立移植瘤动物模型.流式分析显示,体外培养的OCI-Ly3细胞对c-Met、Ki-67和survivin等细胞增殖和凋亡相关标记物具有较高的表达;SCID小鼠皮下接种107 OCILy3细胞,成瘤率87.5%;腹腔接种2×107细胞,成瘤率75%;肿瘤组织符合人DLBCL的组织学特征,且分布弥散,具有较高的侵袭性.成功构建了人ABC型DLBCL的小鼠模型,并对OCI-Ly3细胞的体外培养特性和与肿瘤发生相关的免疫标记物进行了较为系统的检测,为DLBCL的进一步研究提供了参考和工具.
A large diffuse B-cell lymphoma (DLBCL) activated B-cell (ABC) cell line OCI-Ly3 was investigated to explore the biological characteristics of ABC type and tumor-associated molecules. The expression of OCI-Ly3 cells cultured under different conditions was compared and analyzed. The growth morphology and biological characteristics of the cells were observed. The expression of tumor-associated antigen in OCI-Ly3 cells was analyzed. Subcutaneous and abdominal cavity were used. After inoculation, animal models of xenograft tumors were established in SCID mice. Flow cytometry analysis showed that OCI-Ly3 cells cultured in vitro had high expression of c-Met, Ki-67, and survivin-related cell proliferation and apoptosis-related markers. SCID mice were inoculated subcutaneously with 107 OCILy3 cells, the tumor formation rate was 87.5%; 2×107 cells were inoculated intraperitoneally with a tumor formation rate of 75%; tumor tissues were in line with the histological characteristics of human DLBCL, and the distribution was diffuse, with high invasiveness. A mouse model of human ABC DLBCL was constructed, and the in vitro culture characteristics of OCI-Ly3 cells and immune markers associated with tumorigenesis were systematically tested, providing a reference and tool for the further study of DLBCL.
