目的观察黑米皮提取物对人前列腺癌PC-3细胞增殖的抑制作用并探讨其机制。方法用不同浓度的黑米皮提取物(20,150和300μg/ml)处理PC-3细胞,通过MTT法、流式细胞仪、western blot法研究提取物对细胞的影响。结果处理组的细胞增殖率随着提取物作用浓度和时间的增加而降低(P<0.05);与对照组相比,处理组细胞凋亡率增加,差异有统计学意义(P<0.05);细胞周期中,G1期细胞百分比显著增多,S、G2-M期细胞显著减少,细胞被阻滞在G1-S期。与对照组相比,处理组细胞的p-p38、p-JNK的蛋白表达则随着提取物浓度的增加而增强。结论黑米皮提取物能够抑制人前列腺癌PC-3细胞的增殖,改变细胞周期的分布,激活JNK、p38通路,诱导细胞凋亡。
Objective To observe the inhibitory effect of black rice bran extract on the proliferation of human prostate cancer PC-3 cells and to explore its mechanism. Methods PC-3 cells were treated with different concentrations of black rice bran extracts (20, 150 and 300 μg / ml). The effects of extracts on the cells were studied by MTT assay, flow cytometry and western blot. Results Compared with the control group, the cell proliferation rate of the treated group decreased as the concentration and time of the extract increased (P <0.05). Compared with the control group, the apoptosis rate of the treated group increased (P <0.05). In the cell cycle, the percentage of cells in G1 phase increased significantly, the cells in S, G2-M phase decreased significantly, and the cells were arrested in G1-S phase. Compared with the control group, the protein expression of p-p38 and p-JNK in the treated group increased with the increase of the extract concentration. Conclusion Black rice extract can inhibit the proliferation of human prostate cancer PC-3 cells, change the distribution of cell cycle, activate JNK, p38 pathway and induce cell apoptosis.
