目的探讨脾肾阳虚型肠易激综合征(IBS-D)病证结合大鼠模型的建立方法。方法 4周龄雄性SD大鼠38只,随机分为正常组10只、模型组14只和温肾健脾方组14只,采用物理化学刺激共同作用的方法,即全周期(4周)番泻叶灌胃联合2周冰醋酸灌肠+固定器束缚、2周球囊直肠刺激+夹尾刺激制备大鼠模型,造模后温肾健脾方组以13.98 g/(kg·d)温肾健脾方灌胃给药2周,模型组同期予等容量蒸馏水灌胃,正常组无干预。观察大鼠形态学表现和体重;比较排便粒数、粪便含水量、Bristol分型,评估肠道敏感性、肠道动力,并观察肠道黏膜病理组织学。结果与正常组比较,模型组精神萎靡,易激惹,体重无明显变化(P>0.05),排便粒数明显增多(P<0.05),粪便含水量增加(P<0.05),Bristol分型中5、6、7型明显增多,肠道敏感性、肠道动力增加(P<0.05);与模型组比较,温肾健脾方组动作较灵活,偶见烦躁,体重无明显变化(P>0.05),排便粒数下降(P<0.05),粪便含水量减少(P<0.05),Bristol分型向4型转化,肠道敏感性、肠道动力下降(P<0.05)。各组大鼠肠道黏膜病理组织学表现均未见明显异常。结论本物理化学刺激共同作用的造模方法可作为制备脾肾阳虚证IBS-D大鼠模型较理想的方法。
Objective To investigate the establishment of a rat model of IBS-D syndrome with spleen-kidney-yang deficiency syndrome. Methods Thirty-eight male Sprague-Dawley rats of 4 weeks old were randomly divided into normal group (n = 10), model group (n = 14) and Wenshen Jianpi Fang group (n = 14). Physiochemical stimulus The rat model was prepared by intragastric administration of laxative leaves combined with 2-week glacial acetic acid enema + fixator stimulation and balloon-rectal stimulation + tail-clamp stimulation for 2 weeks. Spleen and stomach administration for 2 weeks, the model group during the same period to the same volume of distilled water, normal group without intervention. The morphology and body weight of the rats were observed. The number of defecation particles, water content of feces and Bristol type were compared to evaluate the intestinal sensitivity and intestinal motility. The histopathology of intestinal mucosa was observed. Results Compared with the normal group, the model group was apathetic, irritable and had no significant change in body weight (P> 0.05). The number of defecation increased significantly (P <0.05) and the water content increased (P <0.05) (P <0.05). Compared with the model group, Wenshen Jianpi prescription group was more flexible and occasionally irritable, with no significant change in body weight (P> 0.05). The number of defecation decreased (P <0.05), the water content of feces decreased (P <0.05), Bristol type changed to type 4, the intestinal sensitivity and intestinal motility decreased (P <0.05). The histopathological features of intestinal mucosa of rats in each group showed no obvious abnormalities. Conclusion The physico-chemical stimuli modeling method can be used as an ideal method for the preparation of IBS-D rat model of spleen-kidney-yang deficiency syndrome.
