目的 为用KMB17细胞制备口服脊髓灰质炎减毒活疫苗提供适宜条件。方法 检测不同培养液、不同代次、不同细胞龄和不同病毒感染量等因素对病毒滴度及产量的影响。结果 细胞对脊髓灰质炎病毒的敏感性随细胞龄的增加而降低,培养后24、48、72和96h单个细胞中脊髓灰质炎病毒产量分别为986、745、594和237 Lo-.gTCID50;细胞感染脊髓灰质炎病毒后出现细胞病变时间跨度大于48h,早期从病变细胞释放出的病毒在培养上清中不稳定,感染性滴度每24h降低0.25 LogTCID50以上。结论 KMB17细胞培养脊髓灰质炎病毒的感染性滴度和产毒量受培养液营养成分和病毒接种时细胞龄等条件的影响。
OBJECTIVE: To provide appropriate conditions for preparing live attenuated oral poliomyelitis vaccine with KMB17 cells. Methods The effects of different culture media, different generations, different cell ages and different viral infections on the virus titer and yield were tested. Results The sensitivity of cells to poliovirus decreased with the increase of cell age. The poliovirus yield of single cells at 24, 48, 72 and 96 h after culture was 986, 745, 594 and 237 Lo-, respectively. gTCID50. The time span of cytopathic effect of cells infected with poliovirus was more than 48h. The virus released from diseased cells was unstable in the culture supernatant. The infectious titer was reduced by 0.25 LogTCID50 every 24h. Conclusion Infectious titers and detoxification of KMB17 cell-cultured poliovirus are affected by the nutrient content of the culture medium and the cell age at the time of virus inoculation.
