沙咪珠利对小鼠精子畸形和骨髓细胞微核的影响

  [目的]通过沙咪珠利的小鼠精子畸形和骨髓微核,检测受试品的遗传毒性,为沙咪珠利的进一步开发利用奠定基础.[方法]①小鼠精子畸形试验:60只清洁级昆明小鼠随机分为五组,每组12只.依据沙咪珠利LD50(LD50为5776 mg/kg)及其理化性质,试验时设2500 mg/kg体重(高)、1250mg/kg体重(中)、625 mg/kg体重(低)三个剂量组,阳性对照选用环磷酰胺(40 mg/kg b.w.),阴性对照选用无菌水(0.2 mL/10 g).小鼠连续经口给予受试物5天,从首次染毒计第35天颈椎脱臼处死动物,取双侧附睾制片,每只动物计数1 000个精子中的畸形数,计算畸形率.②微核试验:60只清洁级昆明小鼠随机分为五组,每组12只(雌雄各半,分笼饲养).沙咪珠利设2500 mg/kg体重(高)、1250 mg/kg体重(中)、625mg/kg体重(低)三个剂量组,按0.2 mL/10g体积给小鼠经口染毒受试药物.另分别设一阳性对照组和阴性对照组.采用30h两次给药法,于第二次给药后6h取材、制片,计算微核率.[结论]沙咪珠利各剂量组与阴性对照组比较无显著性差异(P>0.05),与阳性对照组比较,差异有统计学意义(P<0.01).[结论]初步判定沙咪珠利对生殖细胞无遗传毒性.
