Objective: The serum/plasma proteome was explored for biomarkers to improve the diagnostic ability of CA 19-9 in pancreatic cancer (PC).Methods: This study have developed by three study sets 1) training set of 160 serum samples, 2) validation set-1 of 80 plasma samples, and 3) validation-2 of 79 serum sample included PC stage I-IV, disease controls (DC) and healthy volunteers (HV).The blood samples of the training and validation set-1 were analyzed by SELDI-TOF MS.The validation set-2 was analyzed by ELISA Results: A panel ofApolipoprotein (Apo) A-II and Apo C-I were indentified from training set PC vs.DC and from PC vs.HV samples gave a ROC of 0.90 and 0.90 respectively, compared with 0.87 and 0.91 for CA19-9.The ROC AUC was greater (0.97 and 0.99, P<0.05) when CA19-9 was added to the panels and confirmed by validation-1 samples.A combination of ApoA-Ⅱ, ApoC-Ⅰ, and CA 19-9 was tested by the validation-2 using ELISA, where the ROC was greater than that of CA19-9 alone for PC vs.DC (0.90 v.0.84) and for PC vs.HV (0.96 v.0.90).Conclusions: A diagnostic panel ofApoA-II, ApoC-I, and CA19-9 improves the diagnostic ability of CA19-9 alone.This indicates that the levels of these two novel proteins have potential to improve the diagnostic accuracy of CA19-9 sufficiently and potentially contribute to clinical detection at risk patients.
