High salt (HS) intake involves reduced sk current in autonomic paraventricular nucleus neurons (PVN)

  Hypertension induced by Ang Ⅱ and HS (2% NaCl) intake involved reduced SK current of RVLM projecting PVN (PVN-RVLM) neuron.We hypothesized that HS intake alone reduces SK current in PVNRVLM neurons.In whole cell voltage-clamp recording, the anplitude of SK current from the HS (13±5) pA was significantly less than that from normal salt (NS, 0.4% NaCl) (63±11) pA.In whole cell current-clamp recording,graded current injections evoked graded increases in neuronal activity from both groups.Maximum discharge in the HS was (43±4) Hz, which was significantly greater than that of NS.The SK channel blocker apamin significantly increased discharge in NS (47±4) Hz, but had no effect in the HS (45±2) Hz.The contribution of SK channel activity in regulating SNA was assessed by injecting apamin into the PVN of rats with and without HS.Maximum increase in splanchnic SNA and renal SNA by apamin were (319±41)% and (277±48)% in NS, contrasted by a much smaller increase of (116±41)% and (81±28)% (P<0.01) in HS.Our data indicate that reduced SK current in PVN-RVLM neurons contributes to the increased excitability from HS, which may contribute to the sympathetic activation.
