For the past decade,we have developed Tol2-transposon mediated genetic methods,including transgensis,gene trapping,enhancer trapping and the Gal4FF-UAS system in the model vertebrate,zebrafish.By employing these methods,we have performed large-scale genetic screens,and generated more than 1,000 transgenic lines that expressed Gal4FF,an engineered Gal4 transcription activator,in specific cells,tissues,and organs.By crossing these fish with UAS reporter fish,the Gal4FF-expressing cells are visualized,and,by crossing them with appropriate UAS effector fish,functions of Gal4FF-expressing cells can be modified via targeted expression of the effector genes.To make the best use of them,we constructed a web-based database named zTrap(http://kawakami.lab.nig.ac.jp/ztrap/),by which researchers can search transgenic fish of their interests based on expression patterns and genomic loci.Thus,our transgenic fish resource is valuable for developmental biological and biomedical studies.
